Ligand Based Pharmacophore Model Development for the Identification of Novel Anti-Psychotic Drugs

Bipolar disorder (BD) is a severe biological disorder that affecting approximately 1.2% of the adult population. It can be confused with other disorders, including a variety of anxiety disorders and psychotic disorders. Persons with bipolar disorder also suffer repeatedly from psychiatric disorders that are “comorbid” with the bipolar illness. Antipsychotics are recommended for use with the schedules of manic or depression episodes with mental illness, and as likely aides in non-psychotic episodes. Antipsychotic drugs olanzapine and risperidone were generally chosen over conventional antipsychotics. A ligand-based pharmacophore approach has been used for twenty compounds for designing new drugs by using ligand scout software and distance estimation by using Visual Molecular Dynamics (VMD). Four pharmacophoric features i.e. two aromatic ring, one hydrophobic center and one hydrogen bond acceptor feature were identifying for antipsychotic drugs. Pharmacophoric models are then applied on dataset of twenty compounds with known antipsychotic activity. All compounds were filter out by using Pharmacophore based virtual screening. This pharmacophoric model can be further used for identification of novel antipsychotic drugs.
